If you run the mission as a team, each person on your team has to recover one Defixio. However, if you play on the solo mod, you must recover only one Defixio. However, one important thing to remember is your team member can grab a Defixio for you and hand it over to you at the end zone. There is a down volunteer in each section, and once you talk to them, they will tell you where the Defixio is located. Defixios have a red aura and make a noise when you are nearing them.
